Step 6. The final state of the PCR cycle is the extension stage. During this stage, complementary bases are added to the single strand DNA template. As the extension cycle continues, the probe is cleaved. When the upstream primer cleaves the probe from the template strand of DNA, the quencher is separated from the fluorescent reporter. The fluorescent signal given off by the reporter is measured by the instrument.
